Stem Cell-Derived Cell Sheets for Tissue Regeneration

The considerable progress made in the field of stem cell-based therapy has shown its significant potential applications for the regeneration of defective tissues and organs. Although previous studies have yielded promising results, various limitations remain and should be overcome for translating stem cell-based therapies to clinics.

As a possible solution to current bottlenecks, cell sheet engineering (CSE) is an efficient scaffold-free method for harvesting intact cell sheets without the use of proteolytic enzymes and may be able to accelerate the adoption of stem cell-based treatments for damaged tissues and organs regeneration.


Cell Sheet Engineering uses a temperature-responsive polymer-immobilized surface to form unique, scaffold-free cell sheets composed of one or more cell layers maintained with important inter cellular junctions, cell-secreted extracellular matrices, and other important cell surface proteins, which can be achieved by changing the surrounding temperature. These three-dimensional cell sheet-based tissues can be designed for use in clinical applications to target-specific tissue regeneration.

Advancements in Stem Cells and Tissue Engineering

The clinical application of stem cells in tissue engineering and regeneration is becoming more important. Though, its application has been limited by the problems like reproducibility of the stem cells, ethical concerns of harvesting some of these stem cells, and monitoring the fate of stem cells in vitro and in vivo.


The advent of tissue engineering and regeneration has led to the fabrication of advanced biomaterials and scaffolds with enhanced ability to mimic and control the cellular microenvironment like that of innate stem cell niche. Combining the use of stem cells with biomaterials and scaffolds especially synthetic hydrogels that have exhibited physicochemical abilities and properties like native niche can be the upcoming of tissue engineering in terms of formation of new tissues like bones.

Currently, there has a growth in the usage of either endothelial progenitor cells (EPCs), induced pluripotent stem cells (iPSCs), or adult mesenchymal stem cells in preclinical studies: though, this is yet to be transferred to clinical setups as there are limitations in terms of regulations and ethical considerations.

The engineering of human tissues to cure diseases is a branch of science and a very striking field of research together in academia and the biotechnology industrial divisions. Three-dimensional (3D) biomaterial scaffolds can play a critical role in the growth of new tissue morphogenesis through interacting with human cells.

Although simple polymeric bio-materials can deliver mechanical and physical properties required for tissue development, inadequate biomimetic property and lack of interactions with human progenitor cells remain challenging for the promotion of functional tissue formation.



Therefore, the growths of advanced functional biomaterials that respond to stimulus could be the next choice to develop smart 3D biomimetic scaffolds, actively interacting with human stem cells and progenitors along with structural integrity to form functional tissue within a short duration.

Currently smart bio-materials are designed to interact with biological systems for a wide range of biomedical applications, from the distribution of bio-active molecules and cell adhesion mediators to cellular functioning for the engineering of functional tissues to treat diseases.

Stem cells in Tissue Engineering and Regenerative Medicine

With the increasing number of patients suffering from damaged or diseased organs and the shortage of organ donors, the need for methods to construct human tissues outside the body has arisen. Tissue engineering is a newly developing biomedical technology and methodology which combines the disciplines of both the materials and life sciences to replace a diseased or damaged tissue or organ with a living, functional engineered substitute. The so-called triad in tissue engineering encompasses three basic components called scaffold, cell and signalling bio molecule.

Tissue engineering and regenerative medicine not only offer fast recovery but also reduce the medical and economic problem among patients suffering from vascular diseases. Researchers have accumulated a lot of evidence on and developed techniques for bio-fabrication and stem cell biology, but the efficiency of clinical applications of these techniques remains to be clarified.


The use of stem cells holds important promise for the treatment of several injuries and their difficulties. These applications comprise the treatment of injuries to the skin, sensory organs, nervous system tissues, the musculoskeletal system, circulatory/pulmonary tissues and genitals/testicles and of acute radiation syndrome and the development of novel biosensors. The new research developments in these areas suggest that solutions are being developed to reduce critical consequences of wounds and exposures suffered in warfare.

Previously the injuries that would have bring about in deaths but now that results in wounds today; similarly, today’s enduring wounds may be reduced to tomorrow’s bad memories with further advances in stem cell-based therapies.
